I tried to do what wiki suggests and create a campaign.override file with the new directory to where the campaigns are but its not working. How can I change my default campaign directory?

gonna need a little more info than that to be able to assist..

Where is your current in stall located?
Where are your attempting to redirect to?

where is your current campaign.override file located?
what is the contents of your campaign.override?

screenshots might also help of both locations..

    C:\Program Files\MesaMundi\D20PRO\

C:\Users\Wheels\Dropbox\Age of Worms\Online\D20Pro but have tried other locations to test like c:\temp or d:\temp

    C:\Program Files\MesaMundi\D20PRO\judge

single line with C:\Users\Wheels\Dropbox\Age of Worms\Online\D20Pro but have modified to c:\temp or d:\temp for testing

Well, the first thing I would do.. Move both the App and the store location Outside of 'Program Files' and/or 'Users' ...
both can cause annoying access problems due to windows overly aggressive security.. yes, you could fight with windows, set up exceptions and the like, but honestly, it's just easier and more reliable to avoid it altogether.

    here is a local install

here is an example of an override file. Also note, it is using the network drive name, not a mapped drive association.

Also, if you are using notepad, wordpad or the like.. and you have file extensions hidden in wondows.. You may want to make sure that the editor is NOT adding a .txt to the end of the override file.
