   I am looking into buying d20pro and was wondering if it supports the def /DR optional rule set? As opposed to AC .

   You can certainly setup the DR for each creature appropriately, though it doesn't automatically reduce the DR if you are fighting a colossal enemy (I haven't read much of the def/dr rule variant, so if I missed something please post the rule you want to check) - you can do this manually though.
